On my Windows 10, I have default text editor set to Notepad++
But when I click Open button in Spine Settings/Application/Files under Hotkeys section the file gets opened with default Notepad.

It's probably the file association you set on your computer. Mine opens always with Sublime text!
To change it navigate to the folder where the hotkeys are, right-click the hotkeys.txt file, choose "open with" and set a different application.

The .txt
format is already set to open with and it does open with Notepad++ if I locate and double click it. However It opens with Default Notepad when I hit open from settings of spine editor.
Hmm you're right. Even with file associations it will open with Notepad. I'll investigate and see if this is something that can be changed.
Update: In the next update Spine will correctly open with the associated application.
